Dr. Stockwell and his team will develop a humanized, two-enzyme cleavable prodrug system, the activating enzymes of which are produced in situ only when an engineered immune cell engages two tumor antigens. The proposed platform integrates cleavable-linker conjugation chemistry, biochemical logical gates, proximity-based activation, and cell-localized delivery to advance novel chemistry for localized and targeted cancer therapy.
